Effects of Exercise on Mitochondrial Calcium Uniporter and Dysfunction in the Hippocampus of Aged Rats

Abstract
Purpose: This study was to investigate the effects of aerobic exercise on mitochondria dysfunction, mitochondria calcium uniporter (MCU) and apoptosis factors (Bax and Bcl-2) expression in the hippocampus of aged rats. Methods: Thirty-two male Sprague-Dawley rats were randomly divided into four groups: 16 weeks control (16W-CON, n=8), 16 weeks exercise (16W-EXE, n=8), 42 weeks control (42W-CON, n=8), 42 weeks exercise (42W-EXE). The rats in the exercise groups were forced to run on a treadmill for 30 min 4 times per week for 8 weeks. Results: In the present study, aging increased MCU protein expression and induced mitochondrial dysfunction with apoptosis. Compared with the 42W-CON group, in the 42W-EXE group MCU and Bax protein expression were significantly decreased and mitochondrial function showed a significant improvement(p<.05). Conclusion: The findings of the present study indicate that aerobic exercise may assist in preventing overexpression of MUC proteins during brain aging, which leading to alteration of mitochondrial dysfunction and increased apoptosis factors. Therefore, MCU is thought to be a new potential target for the prevention and improvement of neurodegenerative diseases.
